口腔扁平上皮癌の低酸素環境におけるEMTの解明と治療標的としての意義 Significance as a therapeutic target and elucidation of EMT in a low oxygen environment of oral squamous cell carcinoma 1000020404501 石田, 喬之 ISHIDA, Takayuki open access 口腔癌 低酸素 EMT Notch 2011-2013年度科学研究費助成事業（基盤研究（C））研究成果報告書　課題番号：23592968　研究代表者：石田喬之（鹿児島大学・医学部・歯学部附属病院・助教） 口腔癌細胞株を低酸素環境と通常環境で培養し、運動能と浸潤能、E-cadherinの発現を比較した。結果、低酸素環境での亢進を認め、Notch阻害薬を添加することでそれらは抑制された。またNotch阻害薬のまたリアルタイムPCRにてNotch関連因子の発現を調べたところ、低酸素環境でNotch関連因子の発現の亢進を認めた。以上より低酸素環境はNotch経路を介して口腔癌細胞株にEMTを誘導している可能性が示唆され、口腔癌の転移抑制の治療標的になりうることが示唆された。 We compared invasion and motility and expression of E-cadherin in oral cancer cell linescultured in a normal environment and low oxygen environment. They were increased in a low oxygen environment, but suppressed by the addition of Notch inhibitors. We examined the expression of Notch-related factors in the real time PCR, showed an enhanced expression of Notch signaling in a low-oxygen environment.
