Acquisition of T-maze or shuttle box aviodance behaviors was significantly suppresscd in mice,after daily administration of MMC (6mg/kg/day p.o.) for scvcn days. ACh levels in the hippocampus was decreased in mice even 3 hours after single oral dose of MMC (6mg/kg). After 3 days of the treatment with MMC (6mg/kg p.o.,6 successive days),ACh levels in all regions tested (cerebral cortex, cerebellum,hippocampus,corpus striatum, diencephalonmesencephalon and pons medulla)were significantly decreased in mice. In vitro study, ACh release from hippocampal slice of MMC treated (6mg/kg p.o.) mice was significantly enhanced by high potassium stimulation. In rats,daily oral administration of MMC (5mg/kg) for 10 successive days produced significant decrease of ACh levels in 5 regions without pons medulla, and suppressed the retrieval in conditioned avoidance behavior (Sidman type). In vivo microdialysis study,ACh release from rats corpus striatum was significantly enhanced by MMC (10^{-4}M) contained perfusate.
