{"created":"2023-07-25T08:04:19.710528+00:00","id":2502,"links":{},"metadata":{"_buckets":{"deposit":"6ee7a474-b830-4a57-b30a-48bafffc46fa"},"_deposit":{"created_by":18,"id":"2502","owners":[18],"pid":{"revision_id":0,"type":"depid","value":"2502"},"status":"published"},"_oai":{"id":"oai:ir.kagoshima-u.ac.jp:00002502","sets":["99:100"]},"author_link":["13198"],"item_8_date_6":{"attribute_name":"作成日","attribute_value_mlt":[{"subitem_date_issued_datetime":"2012-06-11","subitem_date_issued_type":"Collected"}]},"item_8_description_4":{"attribute_name":"要約(Abstract)","attribute_value_mlt":[{"subitem_description":"2009-2011年度科学研究費助成事業(科学研究費補助金(基盤研究(C))研究成果報告書 課題番号:21591900 研究代表者:米和徳 (鹿児島大学医学部教授)","subitem_description_language":"ja","subitem_description_type":"Other"},{"subitem_description":"本研究では、老化促進マウス(SAM)を用いて脊髄前角細胞の加齢変化と長期的な運動が及ぼす影響を検討した。オートファジーの関与を調べるため、封入体形成因子であるp62とオートファジー実行因子であるBeclin1の発現について検討した。実験動物にはSAMの12週齢(若齢群)、58週齢(老齢群)、50週齢時よりトレッドミル運動(13m/min., 週6日間, 1日2回, 各20分間)を8週間継続して行なった群(老齢運動群)を用いた。脊髄前角細胞数は老齢群で若齢群と運動群よりも有意に減少していた。脊髄前角細胞におけるp62の発現を検討した結果、p62陽性細胞数とp62発現量は3群間に有意差が認められなかった。脊髄前角細胞におけるBeclin1の発現を検討した結果、Beclin1陽性細胞数は、老齢群で若齢群と老齢運動群よりも有意に減少していた。また、Beclin1発現量については、老齢運動群のBeclin1発現量が他の2群に比較して有意に増加していた。老齢群の脊髄前角細胞数は若齢群と老齢運動群に比べて減少していたが、p62陽性細胞数と発現量に有意差は認められなかった。老齢運動群についてはBeclin1陽性細胞数が減少しておらず、発現量は増加していたため、運動により脊髄前角細胞のオートファジー活性が高まることが示唆された。本研究結果から考えると、オートファジーによって変性タンパク質の蓄積を選択的に分解できなくなることが、加齢による脊髄前角細胞の脱落を招くと考えられ、長期的な運動によりオ\nートファジーの機能を高めることが、脊髄前角細胞の脱落を抑制すると推測される。","subitem_description_language":"ja","subitem_description_type":"Other"},{"subitem_description":"The purpose of this study is to examine the effect of treadmill running on age-related\nchanges of spinal motoneurons and to clarify those mechanisms. Forty-three\nsenescence-accelerated mice (SAM) were randomely divided three groups as follows,\nthe young group, the aged group and the exercised group. Twelve young group mice\nwere sacrificed at the age of 12 weeks. Sixteen aged group mice were sacrificed at the\nage of 58 weeks. Fifteen remaining mice were subjected to treadmill running for 8\nweeks from at the age of 50 weeks and were sacrificed at the age of 58 weeks; those\nwere referred to as the aged exercised group. After the sacrifice, spinal cords were\nremoved en bloc and served for histological examination, immunohistological\nexamination and western blotting. The number of motoneurons in aged group was significantly smaller than those in\nother two groups. There is no significantly difference was observed in the number of\np62 positive cells and the amount of p62 expression on western blotting among three\ngroups. The number of Beclin1 positive cells in aged in aged group was significantly\nsmaller than those in other two groups. On western blotting examination, the amount\nof Beclin1 expression in aged exercised group was significantly larger than those in\nother two groups.\nIn this study, number of motoneurons and activity of autophagy were decreased in\naged mice spinal cord. These findings suggest that accelerated activity of autophagy is needed to maintain the intracellular metabolism for prevent neuronal cell death in\naged spinal cord.
