A high-molecular-weight surface protein antigen (PAg) of Streptococcus sobrinus has been given much attention as a candidate of dental caries vaccine. The complete nucleotide sequence of the gene for PAg of Streptococcus sobrinus MT 3791 (serotype g) was determined. The gene consisted of 4,698 bp and encoded a protein of 1,566 amino acid residues. A putative signal peptide was found in the aminoterminal end of the protein. A potential promoter sequence and a putative Shine-Dalgarno sequence preceded the open reading frame. Two internal repeating amino acid sequences were present in the PAg. One repeating region,located in the aminoterminal region,was rich in alanine,and the other,located in the central region,was rich in proline. Membrane-anchor region,located in the carboxyterminal region,was constructed with hydrophobic amino acids.
