The temperature experiments were conducted with 3 different conditions such as 15.0+0.5°C, 23.0+1.5°C and 32.5+0.5°C. Also, 3 grades of the specific gravity with 1.016, 1.024 and 1.032 were prepared. As the light experiments, 3 types of photoperiods, 18L-6D (18hrs. with light and 6hrs. with dark), 6L-18D (6hrs. with light and 18hrs. with dark) and natural photoperiod. were tested. Samplings in the natural ones were taken place at 5:00, 12:00, 17:30 and 23:00 in order to investigate a diurnal rhythm. The examinations were dealt relatively with an application of the quantitative method to the PAS-positive granules observed in the cells of the histological specimens. Effects of the temperature, the photoperiod and the specific gravity were slightly or no exist in the cells. On the contrary, the quantity of the granules showed a feeble diurnal variation.
