| 内容記述 | (Purpose) The effect of CDP-choline intrathecal administralion on Mongolian gerbil's forebrain ischemia was analyzed on the basis of time-courses of blood flow and EEG and of pathological findings. (Methods) First， as a preliminary experiment， the effect of and optimal drug concentration for the above intrathecal administration were examined. As a result. excessive intrathecal administration of CDP-choline was confirmed to cause spasmogenicity，thus 5μl (20mg/kg) was used as dose of administlation. The experiment was performed under spontaneous respiration with a cannula being indwelled intracisternally. While loading with forebrain ischemia for 10 minutes，cerebral blood flow and EEG were recorded， and a group of pre-loading intrathecal administration of physiological saline (n=2) were prepared. For both groups， pathological specimens were prepared after cerebral perfusion and fixation on day 7. (Results) No significant difference in cerebral blood flow and EEG was noted between both groups. Pathohlstologically， the CDP-choling-given group showed a tendency to slighter neurocytonecrosis. (Discussion) CDP-choline has already been shown to be an important co-enzyme for the metabolism of phospholipid， main constitutive component of cell membrane. The recognized effect of CDP-choline adminislered intrathecally can be interpreted as such that the effect of CDP-choline on cell membranous phospholipid causes inhibition of an increase in free fatty acid and of the generation of lipid peroxide and free radical generating in a series of subsequent metabolic processes，on one hand and the retention of mitochondrial function causes inhibition of an increase in intracellular Ca and of subsequent decomposition of structural protein following re-opening on the other. |
