| 内容記述 | 変形性関節症(Osteoarthritis:OA)の病因は未だ不明である。ストレス反応性分子Gadd45・・が初期・・に特徴的に発現していることから、この分子が・・の病態に関与していると仮説し、その遺伝子改変動物・ Gadd45 ベータトランスジェニックマウス)作成を計画した。また、Gadd45・の下流で標的遺伝子を活性化させるC/EBP・のトランスジェニックマウスも作成した。Gadd45・、C/EBP・両分子を、軟骨特異的分子であるCOMP(cartilage oligometric matrix protein) promoter をTet-off system で誘導して発現調節できるdouble transgenic mice として作成した。両マウスの遺伝子発現を確認した。 | |||||

| 内容記述 | Pathology of the osteoarthritis (OA) is uncovered. Gadd45(growth arrest and DNA damaged inducible 45 beta) is a small protein (18kD) which is involved in cell cycle, apoptosis, stress, and hypertrophy. Since Gadd45β is characteristically expressed in clusters of early stage of OA, we hypothesized that Gadd45β is one of the key molecules in the process of OA. This study was designed to establish the conditional transgenic mice. The promoter of COMP was used in Tet-off system. DT mice was produced and the expression of Gadd45βwas confirmed. C/EBPβ, the downstream of Gadd45β, DTmice was also produced and the gene expression was confirmed. | |||||
